Sam Rami is still on board (there was talk of him being off the project).

The villian is going to be kept hush-hush; only the top four/five major characters will have the full script. Others will only get their parts two weeks in advance.

Sam did give this info:

You don't remember that McFarlane comic of Spidey versus The Lizzard ? On two ocations he was denied the final killing of the webhead. I already mentioned that Dr. Connors was named in the first film, and he showed on the 2nd. Lizard can only be CG, as is the transformation.
